How Do We Isolate Exosomes?

Out of many methods standardized for the isolation of exosomes, size-based exclusion of qualified exosomes is found to be the most efficient, safe method for therapeutic applications. We use an advanced ultrafiltration technique to isolate exosomes based on their molecular weight, by choosing suitable membrane filters. The process does not require any special equipment and is less time-consuming, and more cost-effective as compared to other existing methods of isolation of exosomes like ultracentrifugation, tangential flow filtration, etc. The below-mentioned pictorial presentation will help you understand, how easily exosomes are separated from mesenchymal stem cells.

How Exosomes are Characterized at Advancells

It has been proven that the characterization of exosomes isolated from mesenchymal stem cells sourced appropriately is important for early detection, disease monitoring, and the development of effective therapeutics. To better define exosome characterization and facilitate the downstream application, Advancells has formulated a variety of protocols to better define the characteristics of exosomes based on various technologies.

The technologies that we have applied for exosome characterization shall include nanoparticle tracking analysis, FACS sorting, western blotting, and MASS spectrometry (MS) analysis.

One of the well popular methods of counting nanoparticles based on highly sensitive visualization is Nanoparticle Tracking Analysis. The method is based on the principle of direct detection of the light beam that is scattered through an exosome or microvesicles in liquid suspension. The scattered light is collected and detected with the help of a light microscope and then imaged.

A graph mentioning plots of scattered light spots and their speed of motion provides the data that facilitates the determination of total practice count and size distribution.

Exosome Drug Delivery Technology Platform

Exosomes are nanosized information packages released by stem cells and other cells while communicating with each other. These packages are released in extracellular spaces during cellular communication and are absorbed by other cells in a functionally relevant manner. These extracellular vesicles are preloaded with a variety of information including proteins, genetic material, etc. with the ability to hijack other cells and induce them functionally to behave the way is required. Thus, under certain conditions, exosomes generated from stem cells can be manipulated in different ways to carry different cargoes. With this principle, a wide variety of therapeutic molecules, including stem cells can be loaded into exosomes to protect the active molecules from breakdown and to target a specific area of the body. Advancells has therefore developed a technology to produce various active therapeutic components from cultured and the most potent mesenchymal stem cells, at a commercially relevant scale.

The ability to produce commercially valuable therapeutic products from stem cell-derived exosomes demands highly potent stem cells that are appropriate sources, isolated as per regulatory norms, manufactured in GMP grade facility, grown in the serum-free conditioned medium; and already having demonstrated patient safety.   • Exosomes loaded with stem cells are helpful in targeted delivery
  • Exosomes exhibit a strong anti-inflammatory nature due to the presence of a small molecular compound namely curcumin that is known to protect against brain inflammation evident in various neurodevelopmental and neurodegenerative conditions

Exosome Surface Marker-based Characterization Through Flow Analysis

With the help of flow analysis, it is possible to characterize exosomes isolated for the very specific surface proteins; and can provide critical information related to the physiological state of the parental cell. For that matter, it is very essential to characterize isolated exosomes for surface markers to give more reliable and reproducible results.

  • Flow analysis

It is the most common and reliable method to analyze exosomal surface proteins to confirm different chemical and physical characteristics of exosomes in suspension. The principle of this method is based on the light scattering properties of vesicles to analyze different exosomes based on size, once they are tagged with specific fluorescently labeled antibodies. Accordingly, we used three different antibodies, namely CD81, CD9, and CD 63 to confirm their presence.

  • Western blotting

The method is widely used for detecting specific proteins in a given sample. It is based on the semiquantitative analysis of protein based on the size and intensity of color. We used the western blotting technique to identify the presence and expression level of a specific single protein on the surface of exosomes.
